ABSTRACT

Objective:To investigate the effect of Yingyang Gongji Wan (YYGJ) on hepatoma cell line HepG2, and provide evidence for clinical application. Method:YYGJ-contained rats serum was prepared. Then the inhibiory rate of cells was detected by methye thiazolye telrazlium (MTS) method in both YYGJ group and blank group. Apoptosis of HepG2 was detected by TdT-mediated dUT nick-end labeling (TUNEL) method in blank group,YYGJ group, and 5-fluorouracil (5-FU) group. The mRNA expression and protein expression levels of E-cadherin, Vimentin, metalloproteinase-2 (MMP-2) and Smad4 were detected by Real-time quantitative PCR (Real-time PCR) and Western blot respectively. The invasion ability of HepG2 cells was detected by cell migration assay (transwell). Result:YYGJ-contained serum inhibited the proliferation of HepG2 cells in a time and concentration-dependent manner. As compared with blank group, the inhibitory rate was increased in 5%, 10%, and 20% YYGJ-contained serum groups on the third day (PPPPConclusion:YYGJ-contained serum can inhibit the proliferation of HepG2 cells, induce apoptosis, regulate epithelial-mesenchymal transition (EMT)-related E-cadherin, Vimentin, MMP-2 and Smad4 genes and proteins, and decrease tumor invasion ability. The effect was similar to that of 5-fluorouracil. As a unique prescription, YYGJ can be used as a representative for the treatment of coldness and dampness syndrome of primary liver cancer and its anti-cancer mechanism should be further studied.

ABSTRACT

Objective@#To investigate the prevalence of erectile dysfunction (ED) in men with pre-diabetes.@*METHODS@#This study included 500 men with impaired fasting glycaemia (IFG), 500 with impaired glucose tolerance (IGT), and another 500 with normal blood glucose (NBG), all from Lanzhou. We conducted a questionnaire investigation among the subjects using the International Index of Erectile Dysfunction 5 (IIEF-5).@*RESULTS@#The prevalence rates of ED in the IFG, IGT, and NBG groups were 14.8%, 29.2%, and 33.2%, respectively. After controlling for age, nationality, occupation, education, income, obesity, and blood pressure, the incidence rate was markedly higher in the IFG and IGT than in the NBG group (29.2% and 33.2% vs 14.8%, P 0.05).@*CONCLUSIONS@#The prevalence of ED is higher in men with pre-diabetes than in those with normal blood glucose in Lanzhou.

ABSTRACT

Objective To understand the effects of comprehensive community early intervention on the progression of diabetic nephropathy.Methods A total of 86 cases of diabetic with urine micro protein abnormalities were divided into drug intervention group and community comprehensive intervention group with 43 cases in each group.Drug intervention group received losartan tablets only while comprehensive intervention group received diabetic health education,dietary intervention,proper intervention of aerobic exercise and other lifestyles on drug intervention basis.The intervention lasted for 24 weeks.Then the changes of related indexes were observed and compared.Results The difference of indexes between two groups had no statistical significance before the intervention (P >0.05).However,all indexes decreased after the intervention (P <0.05 )and the glycosylated hemoglobin,blood pressure,fasting blood glucose,total cholesterol, serum creatinine values of the comprehensive intervention group were significantly lower than those of drug intervention group (P <0.05 ).Conclusion Comprehensive intervention could better prevent or delay the progression of diabetic nephropathy comparing with drug only intervention.

ABSTRACT

Objective To investigate the silencing effect of siRNA targeting livin gene on the proliferation and apoptosis of U251 cells. Methods Adenoviral Vector of siRNA targeting livin was constructed and transfected into U251 cells. The gene silencing efficiency was monitored by RT-PCR and Western blotting analylis; the proliferation and apoptosis of U251 cells were detected by MTT assays and flow cytometry, respeclwey Mean while, the therapeijlic effect of the recombrnarrt adenoVirus was also observed en tumor-beaeing mice. Resutts The constructed recombinant adenoVirus effeclwely lnhlbited the expreseion of livn at both mRNA and proton leVlls; it also lnhlbited the proeferalion and promoted the apoptds of U251 cells. In addilton, the recombinant adenoVirus effetively inhibited the humor growth in mice and improved the mean survival period of tumor-beaeing mice. Conclusion Sileneing livin gene expression with recombination adenovirus carrying siRNA targeting liven can inhibit the proliferation and promote the apoptosis of U251 cells, which casts new lights on the treatment of glioma.

ABSTRACT

Objective: To evaluate the inhibitory effects of recombinant adenovirus carrying TEM8 (Ad-TEM8) against angiogenesis and growth of mouse glioma G422 cells in vitro and in vivo. Methods: The Ad-TEM8 was constructed and subcutaneously injected to immunize Kunming mice, and then the splenic lymphocytes were collected for co-culture with G422 cells in vitro. The growth inhibition of G422 cells was observed and IFN-γ secretion by the activated lymphocytes were examined by ELISOT method. In addition, the anti-tumor and anti-angiogenesis effects ofAd-TEM8 were observed in Kunming mice carrying G422 xeno grafts. Results: Ad-TEM8 effectively stimulated the generation of cytotoxic Tlymphocytes (CTLs)in Kunming mice and the secretion of IFN-γ. Subcutaneous injection of Ad-TEM8-induced cytotoxicity effectively inhibited in vivo tumor growth and angiogenesis. Conclusion: Ad-TEM8 can induce CTLs in mice, inhibit tumor growth in vitro and in vivo, and disrupt tumor vasculature. The anti-angiogenesis effect might be a mechanism for its in vivo anti-tumor effect; and TEM8 may serve as a novel target for anti-angiogenesis therapy of glioma.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the relationship between chronic prostatitis (CP) and prostatic calculus (PC).</p><p><b>METHODS</b>We used transperineal ultrasonography (TPUS) to detect PC in 500 normal volunteers and 491 CP patients, and divided them into a CP and a CP + PC group according to the ultrasonographic results. Then we analyzed the NIH-CPSI scores, duration of symptoms and white blood cell count in the expressed prostate secretion (ESP).</p><p><b>RESULTS</b>PC was found in 19.8% of the normal controls, 5% (5/100), 12% (12/100), 19% (19/100), 27% (27/100) and 36% (36/100) in the 20-30 yr, 31-40 yr, 41-50 yr, 51-60 yr and > 60 yr groups, respectively. In comparison, PC was detected in 42.2% of the CP patients, 15.8% (12/76), 30.1% (69/215), 55.7% (59/109), 66.2% (43/65) and 82.8% (24/29) in the above five age groups, respectively, with statistically significant differences between the control and CP groups (P < 0.01). The CP and CP + PC groups showed significant differences in the duration of symptoms and white blood cell count in ESP (P < 0.01) but not in CPSI scores (P < 0.05).</p><p><b>CONCLUSION</b>The incidence of PC is higher in CP patients than in healthy men, and it is associated with inflammation, aging and symptom duration, but not with CPSI scores.</p>

ABSTRACT

Benign prostatic hyperplasia (BPH) is a common disease in older men. At present, 5alpha reductase inhibitor-based medication, preferred by most BPH patients as the first-choice therapy, is taking place of traditional transurethral resection of the prostate. This article presents an update of the researches on the treatment of BPH with dutasteride--a novel 5 alpha-reductase inhibitor.
